Preparation of Phosphate Buffer (pH 7.4)

Phosphate Buffer (pH 7.4)

Purpose: Used in biological experiments to maintain pH close to physiological conditions.

Procedure:

  1. Dissolve 8.0 g NaCl, 0.2 g KCl, 1.44 g Na₂HPO₄, and 0.24 g KH₂PO₄ in 800 ml distilled water.

  2. Adjust pH to 7.4 using HCl or NaOH.

  3. Make up to 1000 ml with distilled water.

  4. Store at 4°C if not used immediately.
